Injection of DNA into mouse eggs for the production of transgenic mice or embryos.

The Core Service will inject DNA provided by the investigator into 150 eggs per construct per line. After injection, the eggs are then transferred to the oviduct of three or four pseudopregnant recipients. If a minimum of 2 founders per transgenic construct are not obtained, the facility injects an additional 80 eggs at no extra coast. If after a second injection session there are still no founders, no future microinjections will be performed. A meeting will be held between the investigator, facility director, and technical personnel to discuss future effects. One week after birth, we will cut toes, and send to the investigator who is responsible to do genotyping. Upon 3 weeks after birth, pups are separated and transferred to the investigator.
The PI understands that while we guarantee at least two founders carrying all or part of the transgene, we do not guarantee expression or germline transmission. If the transgene is embryonic lethal, the PI understands that he/she is responsible for payment in full of fees associated with microinjection, and agrees to this by signing the form of the application.
For BAC transgenics, we only guarantee the injection of 200 emboryos; there is no founder guarantee for BAC transenics.

Difficulties in creating transgenic mice
a) The DNA preparation must be pure of any contaminant and sufficiently concentrated.
b) The lethality of embryo death in utero in foster mice may be associated with the type of DNA construct used.
