Injection of embryonic stem cells into blastocysts for the production of chimeric mice.

C57B1/6 X B6D2F1 blastocysts injection to produce knockout/in mice

Embryonic stem cell clones to be provided by the investigator will be microinjected into a minimum 60-80 blastocysts per session. After injection, the blastocysts are transferred into the oviduct of 0.5 day psudopregnant female and allowed to develop to term. Upon weaning 3 weeks after birth, at least 2 chimeric mice are transferred to the investigator. The core facility cannot guarantee germline transmission.

Difficulties in creating chimeric mice
a) A MAP test is required for any new ES cell lines prior to microinjection.
b) On the assigned day, the freshly harvested ES cell sample (without feeder cells) must be delivered to transgenic facility in order to proceed with the injection.
