Overview of Gene Research
Ctnnb1, which encodes β-catenin, is a key regulatory molecule of the Wnt/β-catenin signaling pathway. This pathway is crucial for tissue homeostasis, regulating cell proliferation, differentiation, and function in various biological processes such as organ development [1,2].
Knockout of the intestinal enhancer of Ctnnb1 (ieCtnnb1) led to reduced Ctnnb1 transcription, compromising canonical Wnt signaling in intestinal crypts. Single-cell sequencing showed it altered epithelial compositions and functions of small intestinal crypts. While it hampered epithelial turnovers physiologically, it prevented the occurrence and progression of Wnt/β-catenin-driven colorectal cancers [1].
In conclusion, Ctnnb1 is essential for maintaining normal biological functions through the Wnt/β-catenin signaling pathway. Studies using gene-knockout models, like the ieCtnnb1 knockout mouse model, have revealed its role in intestinal homeostasis and cancer development, providing insights into disease mechanisms and potential therapeutic targets for colorectal cancer [1].
