From reporter insertion to scarless point mutation, we offer two validated recombineering strategies tailored to your research goals.
One-Step BAC Modification
Best for: Reporter insertion or point mutations (with FRT scar)
This method introduces a gene of interest (GOI) or mutation using a homologous recombination strategy with an FRT-flanked drug-selection cassette. After selection, the cassette is excised, leaving behind a single FRT site.
Typical Applications:
  • Insert a reporter (e.g., lacZ, GFP) downstream of a gene promoter
  • Introduce a point mutation when a noncoding “scar” site (e.g., intron) is available

Two-Step BAC Modification
Best for:Precise point mutations without residual sequences
Two rounds of recombination are performed—first replacing the target with a selection cassette, then replacing the cassette with your final edited sequence. This leaves no FRT “scar” behind.
Typical Applications:
  • Mutate a gene with no nearby noncoding region
  • Edit large single-exon genes where scarless precision is essential

Frequently Asked Questions (FAQs)
What information do I need to provide to start a project?
Just describe your intended modification (e.g., “insert GFP after promoter”), and we’ll take care of the strategy design.
Do I need to send my own BAC clone?
Not necessarily. Provide the BAC ID, and we can source it for you. Or send your BAC in E. coli directly.
What’s the typical turnaround time?
Most BAC modifications can be completed within 4–6 weeks, depending on complexity.
How is BAC integrity confirmed after editing?
We verify the modified region by sequencing and use PCR to confirm no large deletions occurred during recombineering.
Can you create transgenic mice using the modified BAC?
