Stage Description Turnaround Time
Knockout Strategy Design Collaborate to design a nuclease-mediated strategy tailored to your specific gene of interest, maximizing on-target activity while minimizing off-target effects. 1-4 days
Nuclease Expression Vector Construction Construct the knockout vector based on client-approved strategy. Where needed, efficacy tests in cell culture can be conducted. 2-3 weeks
Nuclease Injection into Rat Eggs Co-inject ribonucleoprotein (RNP) into fertilized rat eggs, and implant to obtain founders. SD: 8-10 weeks, Long Evans: 10-16 weeks
Founder Screening Screen pups by PCR to identify those with successful knockout modifications. Where needed, Sanger sequencing can be conducted. 1-2 weeks
Breeding Founders to Obtain F1 Breed the founders to wildtype rats and genotype their offspring to ensure transmission of the knockout allele. 12-16 weeks
