Overview of Gene Research
Drd4, encoding the dopamine D4 receptor, is involved in dopamine-related signaling pathways. Dopamine signaling is crucial for various physiological and behavioral processes, including regulation of mood, reward-seeking behavior, and motor control [4].
In acute kidney injury (AKI), Drd4 was found to be decreased in mouse models of ischemia/reperfusion injury or cisplatin treatment. Activation of Drd4 attenuated nephrotoxicity by suppressing oxidative stress and enhancing mitochondrial bioenergetics through down-regulating ROS generation and NOX4 expression. Mechanistically, it reduced the expression of ISG15, suppressing NOX4 ISGylation and enhancing its ubiquitination for degradation [1].
In liver cancer, Drd4 expression is elevated in liver cancer stem cells (LCSCs) and promotes chemo-resistance and CSC-like phenotypes via activating the PI3K/Akt/GSK-3β pathway to stabilize β-catenin and promote its nuclear entry [2].
In myocardial ischemia/reperfusion injury, Drd4 agonist improved cardiac function, reduced heart damage, and apoptosis in cardiomyocytes by promoting GLUT4 translocation depending on the PI3K/AKT pathway [3].
In conclusion, Drd4 plays essential roles in multiple physiological and disease-related processes. Its functions in protecting against AKI, promoting liver cancer stem cell phenotypes, and mitigating myocardial ischemia/reperfusion injury have been revealed through in vivo and in vitro studies, mainly in mouse models. Understanding Drd4 may provide new therapeutic targets for these diseases.
