Overview of Gene Research
Hif1a, also known as hypoxia inducible factor 1 subunit alpha, is a crucial transcription factor responsive to low oxygen availability. It activates the transcription of genes involved in angiogenesis, cell survival, glucose metabolism, and invasion, playing essential roles in mammalian development, physiology, and disease pathogenesis [4,5].
In endothelial cell-specific atg7 knockout (atg7 KO) mice, ablation of Atg7 upregulated Stat1, which suppressed Hif1a expression, inhibiting ischemia-induced angiogenesis, blood flow reperfusion recovery [3]. In colorectal cancer, USP51 forms a positive feed-forward loop with Hif1a, promoting cancer stemness and chemoresistance [2]. In non-small cell lung cancer, lncRNA HIF1A-As2 epigenetically activates MYC, and together they form a double-positive feedback loop to promote cell proliferation and metastasis [1].
In conclusion, Hif1a is essential in regulating hypoxia-related biological processes. Studies using gene knockout models, like atg7 KO mice, have revealed its significance in angiogenesis, cancer stemness, chemoresistance, and tumor cell proliferation and metastasis, providing insights into potential therapeutic strategies for cardiovascular diseases and cancers.
