Overview of Gene Research
Kit, also known as c-kit or CD117, is a proto-oncogene encoding a receptor tyrosine kinase (RTK) [1,3,4]. It responds to stem cell factor (SCF), and its signaling is crucial for cell proliferation, survival, and migration. The activated Kit leads to autophosphorylation and initiates signal transduction, regulating apoptosis, cell differentiation, chemotaxis, and cell adhesion in multiple cell types. It is involved in important physiological processes such as pigmentation, hematopoiesis, gut movement, fertility, and some aspects of the nervous system [1,3,4].
In gastrointestinal stromal tumors (GISTs), about 80-90% of treatment-naive patients harbor primary Kit mutations. These gain-of-function mutations constitutively activate Kit without the need for SCF, contributing to tumor development [2,5]. In mastocytosis, a mutation of the Kit gene, most commonly D816V, is found in almost all cases and is considered a driver lesion [7]. In melanoma, Kit-mutated melanoma represents a rare subset, and activating alterations of Kit are critical to oncogenesis [6].
In conclusion, Kit plays essential roles in various physiological processes through its signaling pathway. Model-based research, especially studies on gene mutations in diseases like GISTs, mastocytosis, and melanoma, has revealed its significance in disease pathogenesis. Understanding Kit's functions and its dysregulation in diseases provides potential targets for therapeutic intervention.
