Lmna, which encodes lamin A/C, is a gene crucial for maintaining nuclear mechanostability and function, chromatin organization, and gene transcription as a component of the nuclear envelope [1]. Mutations in Lmna can lead to a variety of diseases known as laminopathies, including LMNA-related cardiomyopathy, a common and highly penetrant inherited heart disease with a poor prognosis [1].
Mouse models have been used to characterize the phenotypic diversity caused by specific Lmna variants and understand the pathogenesis of heart disease. These models have shown that Lmna mutations disrupt nuclear envelope stability, activate the DNA damage response, and compromise chromatin organization and mechanotransduction, leading to fibrosis, arrhythmias, and myocardial remodeling [2].
In conclusion, Lmna is essential for normal nuclear function and its disruption through mutations can lead to severe cardiac phenotypes.
