Overview of Gene Research
Parvb, also known as beta-parvin, is an actin-binding and focal adhesion protein. It is involved in linking integrins and associated proteins with intracellular pathways that regulate actin cytoskeletal dynamics and cell survival. The ILK-β-parvin pathway, where Parvb is a part, can enhance cell motility [2].
In a mouse model (PARVB cKO) where the Parvb gene was specifically deleted from proximal tubular epithelial cells, depletion of Parvb significantly attenuated cisplatin-induced renal tubular injury, including tubular cell death and inflammation. Mechanistically, Parvb associates with TAK1, and depletion of Parvb promotes cisplatin-induced TAK1 degradation, inhibits TAK1 downstream signaling, and alleviates cisplatin-induced tubular cell damage [1].
In conclusion, Parvb plays a crucial role in cell adhesion, spreading, motility, and survival. The study of Parvb cKO mouse models reveals its significance in cisplatin-induced renal tubular injury, suggesting that targeting the Parvb-TAK1 pathway may offer a novel therapeutic strategy for alleviating cisplatin-induced kidney damage.
