Lypla1, also known as acyl protein thioesterase 1 (APT1), is an enzyme involved in protein depalmitoylation, the process of removing thioester-linked long-chain fatty acids from cysteine residues in proteins [2]. It plays a role in balancing the palmitoylation cycle, which is crucial for efficient membrane targeting of proteins, and may be involved in signaling, growth, and cell organization [2].
In the context of hypoxia-induced mitophagy in triple-negative breast cancer (TNBC), Lypla1 was found to depalmitoylate GPCPD1, a key enzyme in choline metabolism. This depalmitoylation facilitated GPCPD1's relocation to the outer mitochondrial membrane, which then bound to VDAC1, interfering with its oligomerization. The increased monomer of VDAC1 provided more anchor sites for PRKN-mediated polyubiquitination, ultimately triggering mitophagy. GPCPD1-mediated mitophagy promoted tumor growth and metastasis in TNBC both in vitro and in vivo [1].
Additionally, in the regulation of Porcine reproductive and respiratory syndrome virus (PRRSV-2) replication, LYPLA1 de-palmitoylates the N protein of the virus, counteracting the activity of ZDHHC3 (a palmitoyl transferase) on PRRSV replication. Administration of a small-molecule inhibitor targeting LYPLA1 could hinder PRRSV-2 replication [3].
Also, in the regulation of SQSTM1-mediated selective autophagy, LYPLA1 reverses the S-acylation of SQSTM1 catalyzed by ZDHHC19. S-acylation of SQSTM1 enhances its affinity with the phagophore membrane, promoting efficient autophagic degradation of ubiquitinated substrates [4].
In summary, Lypla1 is essential in protein depalmitoylation processes, which are involved in multiple biological functions such as mitophagy, virus replication regulation, and selective autophagy. These findings from in vitro and in vivo studies highlight its significance in diseases like TNBC and in understanding virus-host interactions. The use of inhibitors targeting Lypla1, as demonstrated in the PRRSV-2 study, also shows potential for therapeutic strategies [1,3,4].
