Rab3a, a small GTP-binding protein, is a member of the Rab family implicated in intracellular vesicle trafficking. In plasma membrane repair, Rab3a is essential for lysosome exocytosis. It forms a tripartite complex with synaptotagmin-like protein 4a and non-muscle myosin heavy chain IIA, which is vital for positioning peripheral lysosomes for repair [1]. In hippocampal neurons, Rab3a interacts with spastin, regulating its protein stability and microtubule-severing function to modulate neurite outgrowth [2]. In cardiomyocytes, zDHHC9 palmitoylates Rab3gap1, influencing Rab3a activity and atrial natriuretic peptide secretion [3]. Silencing the Rab3A/Rab27A system in podocytes affects differentiation, apoptosis, and cytoskeletal organization, highlighting its role in podocyte injury [5]. In a rat spinal cord injury model, Rab3a promoted the secretion of neuronal cell-derived extracellular vesicles by interacting with STXBP1, which inhibited M1 macrophage polarization and thus promoted spinal cord injury repair [6]. In melanocytes, RAB3A regulates melanin exocytosis and transfer induced by keratinocyte-conditioned medium [7]. In murine leukemia virus infection, Rab3a is required for the stabilization of the Gag protein [8]. Heterozygous RAB3A variants, like p.Arg83Trp, cause cerebellar ataxia through a partial loss-of-function mechanism [9].
