Overview of Gene Research
Rps14, the ribosomal protein S14, is a component of the 40S ribosomal subunit and is crucial for ribosomal biogenesis. It may be involved in the formation of pre-RNA 18s, an intermediate for the 40S small ribosomal subunit. Rps14 is associated with multiple biological processes and signaling pathways, such as p53 and AMPK signaling [2].
Rps14 haploinsufficiency in mouse models recapitulates some features of the 5q-syndrome, like more severe anemia and megakaryocyte morphology changes. Macrophages in these mice show activation of the innate immune response [3]. In glioma, loss-of-function experiments revealed that Rps14 depletion suppresses cell proliferation, migration and promotes apoptosis, with its effects closely related to the p53 signaling pathway [1]. In ER-positive breast cancer cells, downregulation of Rps14 inhibits cell proliferation, cycle, and metastasis, and induces apoptosis [4].
In conclusion, Rps14 plays essential roles in ribosomal biogenesis and is involved in various biological processes and disease conditions. Mouse models with Rps14 haploinsufficiency have been valuable in understanding its role in the 5q-syndrome. Studies in glioma and breast cancer through loss-of-function experiments have further elucidated its role in tumor-related processes, highlighting its potential as a therapeutic target in these diseases.
