Overview of Gene Research
Sh2d1a, also known as SAP (SLAM-associated protein), encodes an intracellular adaptor molecule. It is predominantly expressed in T cells and NK cells and functions to regulate signal transduction pathways downstream of the SLAM family of surface receptors. This regulation controls CD4+ T cell (and by extension B cells), CD8+ T cell and NK cell function, as well as the development of NKT cells. The study of genetic models, like gene-knockout (KO) mouse models, can help to reveal its functions in vivo [2].
In KO mouse models, deficiency in Sh2d1a protects mice from an experimental model of lupus, as it impairs T-dependent antibody responses and germinal center formation, while not grossly affecting T or B cell function per se.
