Snapin, a SNARE-associated protein, is involved in multiple essential biological functions. It plays a crucial role in the formation of neuronal synapses, which is vital for the normal growth and development of synapses, thus contributing to brain homeostasis [2]. It also participates in processes related to cell cycle regulation, insulin secretion, and Ca²⁺ channel regulation [1,3].
In pancreatic β cells, Snapin knockdown arrests the cell cycle in the S phase and inhibits cell proliferation, while its overexpression has the opposite effect. This indicates that Snapin mediates β cell proliferation and insulin secretion, suggesting it could be a pharmacotherapeutic target for diabetes mellitus [1].
In neurons, snapin knockout mice exhibit aberrant accumulation of immature lysosomes, impaired retrograde transport of late endosomes, and reduced neuron viability. This reveals that Snapin-DIC coordinates retrograde transport and late endosomal-lysosomal trafficking critical for autophagy-lysosomal function and neuronal homeostasis [4]. Additionally, snapin-deficient cortical neurons show reduced frequency of miniature excitatory postsynaptic currents, smaller release-ready vesicle pool size, and "desynchronized" synaptic vesicle fusion, impairing synaptic precision and efficacy, highlighting its role in synaptic function [5].
