Overview of Gene Research
ERICH3, also known as Glutamate Rich 3, is a gene with significant functions in the body. It has been implicated in vesicular function, especially in serotonergic and other neuronal cell types. This function might be related to antidepressant treatment response. Additionally, ERICH3 is a component of the mammalian primary cilium, regulating cilium length and the ciliary levels of sonic hedgehog signaling molecules [1,2].
In terms of its role in diseases, several studies have focused on its association with major depressive disorder (MDD). Meta-analysis showed that certain SNPs across the ERICH3 gene were significantly associated with SSRI treatment outcomes in MDD patients. ERICH3 may be involved in vesicular function related to serotonin (5-HT), as its knockout in SK-N-SH cells decreased 5-HT staining, and concentrations in the culture media and cell lysates without changing 5-HT synthesizing or metabolizing enzymes. Also, the ERICH3 rs11580409 polymorphism was associated with 6-month antidepressant response in depressed patients [1,3].
In conclusion, ERICH3 plays important roles in vesicular function and primary cilium regulation. Its association with antidepressant treatment response in MDD patients, especially through its potential influence on 5-HT-related vesicular function, makes it a gene of interest in understanding the mechanisms of MDD and developing better treatment strategies.
