Overview of Gene Research
Tcf7, also known as Transcription Factor 7, is a gene expressed in immune cells and is involved in multiple biological processes. It is the key downstream functional molecule of the Wnt/β -catenin signaling pathway, which participates in regulating various cellular activities [7].

In cancer research, TCF7-positive T cells in oral cancer are associated with tertiary lymphoid structures/organs and a superior prognosis. These cells express high levels of TLS-related genes and low levels of immune checkpoint molecules, suggesting they could be a new therapeutic target and prognostic marker [1]. In melanoma, the visualization of TCF7 within CD8+ T cells in fixed tumor samples predicted positive clinical outcome in checkpoint-treated patients [4]. In colorectal cancer, Dihydroartemisinin inhibits cancer development through the GSK-3β/TCF7/MMP9 pathway [6]. Also, in the context of ligamentum flavum hypertrophy, TCF7 promotes the hyper-proliferation and fibrosis phenotype through the TCF7/SNAI2/miR-4306 feedback loop, and its inhibition could be a novel therapeutic approach [7].

In CD8+ T cell research, Tcf7hi effector cells can give rise to central memory CD8+ T cells, and Tcf1 (a product of Tcf7) counteracts the differentiation of these cells [2]. Additionally, a microbial metabolite, indole-3-propionic acid (IPA), enhances immunotherapy efficacy by modulating T cell stemness through increasing H3K27 acetylation at the super-enhancer region of Tcf7 [3].

In the study of glucose homeostasis, Tcf7 expression is not a critical determinant in mice, and its expression in islet mRNA is due to islet-associated murine immune cells rather than islet β-cells [5].

In conclusion, Tcf7 is involved in multiple biological processes, especially in immune-related functions and cancer-associated mechanisms. Research using various models, though not always KO/CKO mouse models, has revealed its importance in cancer prognosis, immunotherapy response, and cell-fate determination in T cells. Its role in non-cancerous conditions like glucose homeostasis seems to be negligible. The study of Tcf7 provides valuable insights into immune-mediated diseases and cancer immunotherapy [1-7].
