Overview of Gene Research
Dnajc27, a member of the HSP40 protein family, was previously identified as a body mass index (BMI) associated locus in genome-wide association (GWAS) studies [3,7]. It is involved in heat shock response, an essential cellular stress response. Dysregulation of heat shock proteins like Dnajc27 within this pathway may contribute to obesity-induced insulin resistance and type 2 diabetes (T2D) [3]. Also, it has been associated with histone chaperoning and ciliogenesis [5].
Integrated analysis of Mendelian Randomization and Bayesian colocalization has revealed that Dnajc27 is among the risk genes associated with a shared genetic basis between inflammatory bowel disease and psoriasis [1]. In a Kuwaiti cohort, the MC4R variant rs17782313 was associated with increased levels of Dnajc27, which in turn reduced the MC4R-mediated formation of cAMP, potentially enhancing appetite and regulating obesity [2]. Moreover, in a Chinese Han preschool children study, the polymorphism at rs713586 locus of Dnajc27 was associated with susceptibility to overweight and obesity [4]. Additionally, Dnajc27 was suggestively associated with Parkinson's disease in a large case-control study [6].
In conclusion, Dnajc27 plays a role in multiple biological processes and disease conditions. Its association with obesity-related traits, inflammatory bowel disease, psoriasis, and potential link to Parkinson's disease highlight its significance in understanding the pathophysiology of these disorders.
