Overview of Gene Research
Tnfsf9, also known as CD137L/4-1BBL, is a member of the tumor necrosis factor superfamily. It functions as a T-cell co-stimulator, playing a role in cancer immunotherapy and being involved in regulating immune cell functions. It has implications in various biological processes related to the immune response and cancer [1,7].

In pancreatic cancer, Tnfsf9 expression is elevated and is associated with poor prognosis. It can promote pancreatic cancer metastasis by inducing M2 macrophage polarization through Src/FAK/p-Akt/IL-1β signaling [3], as well as through Wnt/Snail signaling [4].

In glioma, hypoxia can upregulate Tnfsf9 expression in a histone-lactylation-dependent manner, leading to M2 macrophage polarization and promoting glioma progression [2].

In hepatocellular carcinoma, Tnfsf9 expression is downregulated, and overexpression or treatment with its recombinant protein can inhibit HCC cell proliferation, migration, and invasion both in vitro and in vivo, suggesting it may be a tumor suppressor [7].

In patients with metastatic renal cell carcinoma treated with nivolumab and ipilimumab, Tnfsf9 expression may predict treatment response, with high-expression tumors showing an activated adaptive immune response [5].

Inherited Tnfsf9 deficiency in a patient led to broad Epstein-Barr virus infection with EBV+ smooth muscle tumors, highlighting the role of the CD137-CD137L (Tnfsf9) pathway in anti-EBV immunity [6].

In conclusion, Tnfsf9 has diverse functions in the immune response and cancer. Its role in promoting or inhibiting tumor progression varies depending on the cancer type. Studies on Tnfsf9, including those using relevant models, have provided insights into its functions in diseases such as cancer and viral infections, which may help develop new therapeutic strategies.
