Overview of Gene Research
Wt1, also known as Wilms' tumor 1, encodes a zinc finger nuclear transcription factor and RNA-binding protein. It is crucial for the development of many organs, playing roles in cell growth, differentiation, and lineage specification. In the kidney, it is involved in nephrogenesis, and in gonads, it is essential for the survival of genital ridge cells [2,3].
In a study using ischemia-reperfusion injury (IRI) mouse model, conditional deletion of Wt1 gene in proximal tubular epithelial cells (PTECs) and glomeruli parietal epithelial cells (PECs) aggravated renal tubular injury after severe acute kidney injury. This reveals that Wt1 in PECs and PTECs is essential for renal proximal tubule regeneration after severe kidney injury [1]. In another mouse model study, homozygous mutation of Wt1 led to gonad agenesis, indicating its requirement for the survival of genital ridge cells [2].
In conclusion, Wt1 is vital for organ development, especially in the kidney and gonads. Gene knockout and conditional knockout mouse models have shown its significance in renal proximal tubule regeneration after acute kidney injury and in gonad development. Understanding Wt1's functions through these models provides insights into the mechanisms of organogenesis and disease, potentially guiding future therapeutic strategies for related diseases.
