Pgm5, phosphoglucomutase-like protein 5, is a metabolic enzyme involved in the metabolism of glucose-1-phosphate and glucose-6-phosphate [1,2]. Although its exact function and associated pathways are still being explored, metabolic enzymes like Pgm5 are crucial for maintaining normal cellular metabolic processes, which are essential for cell survival, growth, and differentiation. Genetic models, such as gene knockout (KO) or conditional knockout (CKO) mouse models, could potentially be valuable in further understanding its function.
Multiple studies have explored Pgm5's role in various cancers. In liver cancer, PGM5 expression was significantly lower in cancerous than adjacent normal liver tissues, and had modest diagnostic value. Its expression had positive associations with male sex and survival, but negative associations with advanced disease features. Low PGM5 levels were associated with poorer overall survival and relapse-free survival, indicating it may be a diagnostic and prognostic biomarker [1].
In colorectal cancer, PGM5 protein levels were lower in cancer tissues, and low PGM5 expression was associated with poor overall survival. In prostate cancer, PGM5 expression was significantly lower in cancer tissue, and its low expression was linked to poor clinical outcome and high Gleason score. Overexpression of PGM5 repressed prostate cancer cell proliferation and migration [3].
In conclusion, Pgm5 appears to play a role in cancer development and prognosis across multiple cancer types. Studies in these cancer models suggest that Pgm5 may act as a suppressor of tumor cell proliferation, invasion, and migration.
