Overview of Gene Research
Gca, also known as grancalcin, is a protein whose levels in circulation are associated with cognitive function. Bone marrow-derived GCA+ immune cells, which secrete Gca, play a role in the pathogenesis of Alzheimer's disease (AD). These cells preferentially invade the hippocampus and cortex of AD mouse models in a C-C Motif Chemokine Receptor 10 (CCR10)-dependent manner [1].
In AD research, transplanting GCA+ immune cells or injecting recombinant GCA protein intensifies amyloid plaque load and aggravates cognitive and memory impairments. Conversely, genetic ablation of GCA in the hematopoietic compartment improves cognitive and memory function. Mechanistically, GCA competitively binds to the low-density lipoprotein receptor-related protein 1 (LRP1) in microglia, inhibiting phagocytosis and clearance of Aβ and potentiating neuropathological changes. GCA-neutralizing antibody treatment rejuvenates cognitive and memory function and constrains AD progression [1].
In conclusion, Gca-related research, especially through genetic ablation models, reveals that Gca+ immune cells play a pathological role in cognitive and memory decline in AD. Understanding Gca's function provides potential targets for innovative therapeutic strategies in AD.
