Overview of Gene Research
Pdk1, also known as 3-phosphoinositide-dependent protein kinase 1, is a master kinase regulating AGC kinase family members. It plays a crucial role in the PI3K/Akt, Ras/MAPK and Myc signaling pathways [1,2]. It is involved in multiple biological processes such as cell growth, metastasis, invasion, apoptosis and survival, making it of great biological importance [1]. Genetic mouse models are valuable tools for studying Pdk1.
In a genetic mouse model, Smurf1 deficiency dramatically reduced colorectal cancer tumorigenesis, unveiling the role of Smurf1-mediated PDK1 neddylation in PI3K-Akt signaling and tumorigenesis [3]. In breast cancer, PDK1 knockdown reduced the proliferation, migration, and tumorigenicity of breast cancer cells and inhibited the HIF-1α pathway [4]. Also, downregulation of PDK1 levels inhibited migration and experimental metastasis of human breast cancer cells [5].
In conclusion, Pdk1 is essential in regulating cell behaviors and signaling pathways. Mouse models have revealed its significant roles in cancer development, including colorectal and breast cancer. Targeting Pdk1 may be a potential strategy for cancer therapy.
