Overview of Gene Research
Tnpo1, also known as Transportin-1 and Karyopherin-β2, is a crucial member of the karyopherin-β family. It is mainly involved in nuclear import processes, facilitating the transport of specific proteins into the nucleus. This function is essential for various cellular processes and signaling pathways, such as those related to cell growth, differentiation, and immune response [1,3,4,5].

In cancer research, TNPO1 has been found to play oncogenic roles. In ARID1A-deficient gynecologic cancer, TNPO1-mediated nuclear import of ARID1B promotes tumor growth. Knocking down TNPO1 or ARID1B changes chromatin accessibility, leading to loss of certain histone markers, diminishing activated transcription factors of the AP-1 family, and inactivating the PI3K/AKT signaling pathway [1]. In esophageal cancer, circBCAR3 can upregulate TNPO1 by interacting with miR-27a-3p, accelerating tumorigenesis and metastasis [2]. In cervical cancer, TNPO1-mediated nuclear import of FUBP1 contributes to tumor immune evasion by increasing NRP1 expression [3].

In conclusion, TNPO1 is vital for protein nuclear import, significantly influencing cellular functions. Its role in cancer, as revealed by functional studies in relevant models, shows its potential as a therapeutic target in treating ARID1A-deficient gynecologic cancer, esophageal cancer, and cervical cancer. Understanding TNPO1 provides insights into cancer mechanisms and potential treatment strategies.
