Overview of Gene Research
Tlr2, Toll-like receptor 2, is a key component in the innate immune system. It functions as a cellular sensor for both microbial pathogen-associated molecular patterns (PAMPs) like those from Gram-positive and Gram-negative bacteria, mycobacteria, spirochetes, and mycoplasm, and endogenous damage-associated molecular patterns (DAMPs) such as heat shock protein (HSP) 60 [5]. TLR2 can form homodimers or heterodimers with TLR1 and TLR6, and is crucial in initiating innate immune responses, which may involve oxidative stress and cellular necrosis initially spurred by microbial compounds [5].

TLR2 plays diverse roles in various diseases. In hepatitis B, it may participate in both the immune response against HBV and the induction of virus-related hepatic complications like hepatocellular carcinoma (HCC) and liver cirrhosis, though recent studies suggest it can also fight against HCC and liver cirrhosis, with factors like nutrition habits, gene polymorphisms, gut microbiome, HBV antigens, and other receptor activation influencing its functions [1]. In multiple sclerosis, it's hypothesized that a "defective" microbiome fails to provide normal levels of "TLR2-tolerizing" bacterial products, leading to abnormally regulated systemic innate immune TLR2 responses involved in both inflammation and defective remyelination, and induction of TLR2 tolerance may be a novel treatment approach [3]. In retinal diseases such as age-related macular degeneration (AMD) and diabetic retinopathy (DR), dysregulation of TLR2 signalling can lead to an overactive innate immune response, excessive pro-inflammatory cytokine production, and chronic inflammation [4]. In hair follicle biology, TLR2 is expressed in a cycle-dependent manner in healthy hair follicles, governing hair follicle stem cell (HFSC) activation by countering inhibitory BMP signalling, and its decrease in aging and obesity is associated with reduced hair regeneration capacity, while administration of its endogenous ligand carboxyethylpyrrole (CEP) can stimulate hair regeneration through a TLR2-dependent mechanism [2].

In conclusion, Tlr2 is a vital gene in the innate immune system, with its functions extending to multiple disease areas including hepatitis B, multiple sclerosis, retinal diseases, and hair follicle-related conditions.
