Overview of Gene Research
MTNR1B, the gene encoding melatonin receptor 1B, is crucial in various biological processes. Melatonin, a neurohormone involved in circadian rhythms, binds to MTNR1B, and this interaction has implications in multiple pathways. Genetic variations in MTNR1B have been linked to glucose metabolism-related disorders, suggesting its importance in metabolic regulation [3,4].
In a Mtnr1b knockout mouse model, deletion of Mtnr1b disrupted placental angiogenesis via the VEGF signaling pathway, leading to fetal growth restriction. Specifically, Mtnr1b knockout increased placental oxidative stress and apoptosis, reduced angiogenesis and placental vascular density, and these changes were mediated by the disrupted STAT3/VEGFR2/PI3K/AKT pathway [1].
In another study, astrocyte-specific knockout of Mtnr1b in mice (Mtnr1b cKO𝐺𝑓𝑎𝑝 mice) resulted in anxiety-like behavior, caused by down-regulation of gamma-aminobutyric acid-ergic (GABAergic) synaptic function [2].
In conclusion, MTNR1B plays essential roles in placental function and neural behavior regulation. Mouse models with Mtnr1b knockout or conditional knockout have provided valuable insights into its functions in processes such as placental angiogenesis and neural synaptic function, which are relevant to diseases like fetal growth restriction and potentially neuropsychiatric disorders.
