Overview of Gene Research
Gpr101, an orphan G protein-coupled receptor belonging to Family A rhodopsin-like family, is predominantly expressed in the hypothalamus and pituitary. It plays a crucial role in regulating functions like energy balance and reproduction. Gpr101 couples to Gαs and Gαi proteins and is involved in the hypothalamus-pituitary-gonad axis. Its over-expression is linked to X-linked acrogigantism (XLAG) and acromegaly [1].
In transgenic mice with elevated somatotrope Gpr101 expression (GhrhrGpr101), Gpr101 causes elevated GH/prolactin secretion but without hyperplasia/tumorigenesis. It constitutively activates Gs, Gq/11, and G12/13, leading to GH secretion but not cell proliferation, similar to the signatures in human pituitary tumors with high Gpr101 expression [2].
In conclusion, Gpr101 is essential in regulating the somatotrope axis function. The study of Gpr101 transgenic mouse models has revealed its role in growth hormone hypersecretion and provided insights into the pathogenesis of XLAG and related pituitary diseases, contributing to a better understanding of these disease mechanisms.
