Overview of Gene Research
Ube2k, a ubiquitin-conjugating enzyme, is involved in the ubiquitin-proteasome system, responsible for numerous cellular physiological activities. It participates in the synthesis of K48-linked ubiquitin chains, which target substrates for proteasomal degradation [5]. This process is crucial for maintaining normal cellular function, regulating protein levels, and controlling various biological pathways.
In the context of diseases, in hepatocellular carcinoma (HCC), Ube2k expression is elevated, promoting cell proliferation, migration, and stemness property, and a worse prognosis for patients. Hypoxia can induce HIF-1α-mediated increase of Ube2k levels in HCC cells, and Ube2k cooperates with HIF-1α to form a functional axis to drive HCC progression [2,3].
In pancreatic ductal adenocarcinoma (PDAC), Ube2k is highly expressed, promoting cell proliferation and the stemness phenotype both in vitro and in vivo, and IGF2BP3 increases Ube2k expression by enhancing its RNA stability [4].
In glioma cells, Ube2k is highly expressed, and silencing it inhibits cell growth by inducing autophagy-related apoptosis [8].
In multiple myeloma, ixazomib, a 20S proteasome inhibitor, can inhibit cell proliferation by targeting Ube2k [7].
In depression, microglial circ-UBE2K binds to HNRNPU to upregulate Ube2k expression, leading to abnormal microglial activation, neuroinflammation, and promoting the development of major depressive disorder (MDD) [1].
In Alzheimer's disease, elevated Ube2k induced synaptic dysfunction through ubiquitinating K1082 on GluN2B [6].
In conclusion, Ube2k plays a significant role in multiple biological processes and disease conditions. Its involvement in cancer progression, neurodegenerative diseases like Alzheimer's, and mental disorders such as depression indicates its potential as a therapeutic target. Functional studies, including those using gene knockout or conditional knockout mouse models (if applicable in the future), will further enhance our understanding of Ube2k's role in these diseases, providing new strategies for diagnosis and treatment.
