Overview of Gene Research
Sln, also known as sarcolipin, is a protein-related gene. It uncouples sarco -/endoplasmic reticulum Ca2+-ATPase (SERCA), increasing ATP consumption and contributing to heat liberation, thus playing a role in maintaining metabolic homeostasis in human skeletal muscle [1]. This thermogenic mechanism in skeletal muscle may offer a strategy against obesity and related metabolic disorders.
In a study on human skeletal muscle cells with stable Sln knockdown (SLN-KD), glucose and oleic acid (OA) oxidation were diminished, basal respiration was reduced, and the gene expression levels of peroxisome proliferator-activated receptor γ coactivator 1α (PGC1α) and forkhead box O1 (FOXO1) were decreased. Additionally, there was increased lipid formation and accumulation in SLN-KD cells [1].
In conclusion, Sln is crucial for metabolic homeostasis in human skeletal muscle. Findings from Sln-KD cell models suggest that therapeutic strategies involving Sln-mediated futile cycling of SERCA could have significant implications for treating obesity and associated metabolic disorders.
