TRMT6, short for tRNA methyltransferase 6, is an enzyme that catalyzes N1-methyladenosine (m1A), a reversible modification in various RNAs such as tRNA, mRNA, rRNA, and lncRNA. This modification is crucial as it participates in regulating RNA processing, structure, and functions. The TRMT6-TRMT61A complex, in particular, is involved in the m1A methylation of tRNA, which impacts diverse biological processes and is associated with multiple disease-related pathways [1,2,4,5,6,7,9].
In aged murine hematopoietic stem cells (HSCs), an increase in the TRMT6-TRMT61A complex due to declined CRL4DCAF1-mediated ubiquitination degradation signaling impairs HSCs through 3'-tiRNA-Leu-CAG and the RIPK1-RIPK3-MLKL-mediated necroptosis cascade [2]. In hepatocellular carcinoma (HCC), TRMT6 is overexpressed, promoting cell proliferation both in vivo and in vitro through the PI3K/AKT/mTOR axis [3]. Also, in HCC, TRMT6/TRMT61A-mediated m1A methylation in tRNA drives tumourigenesis by regulating cholesterol metabolism [4]. In bladder cancer, depletion of TRMT6/TRMT61A reduces proliferation and resistance against cellular stress [7]. In addition, the rs236110 C > A polymorphism in the TRMT6 gene increases the risk of Wilms tumor [8].
In conclusion, TRMT6, mainly as part of the TRMT6-TRMT61A complex, plays a significant role in m1A methylation of tRNA, influencing diverse biological processes. Model-based research, especially studies using KO/CKO mouse models, has revealed its importance in diseases like HSC aging-related impairments, hepatocellular carcinoma, bladder cancer, and Wilms tumor, providing insights into disease mechanisms and potential therapeutic targets.
