Overview of Gene Research
Mtdh, also known as AEG-1 and LYRIC, is a 582-amino-acid transmembrane protein encoded by a gene at chromosome 8q22. It is distributed in the cytoplasm, peri-nuclear region, nucleus, nucleolus, and endoplasmic reticulum. Mtdh is involved in multiple signaling pathways such as PI3K/Akt, NF-κB, Wnt/β-catenin, and MAPK, and is crucial in various biological processes including cell proliferation, invasion, angiogenesis, metastasis, and chemoresistance in cancer [2,3,4].
Genetic ablation of Mtdh in mice has shown that it inhibits breast cancer development by disrupting its interaction with SND1, which is required for breast cancer progression [1]. Mtdh -/- mice are also protected against dextran sodium sulphate-induced colitis symptoms, indicating Mtdh's role in promoting colitis by facilitating TLR-induced pro-inflammatory cytokine production in colonic lamina propria macrophages [5].
In conclusion, Mtdh is a key regulator in cancer progression and intestinal inflammation. Gene knockout mouse models have been instrumental in revealing its role in breast cancer and colitis, providing potential therapeutic targets for these diseases.
