Overview of Gene Research
Ube2r2, part of the ubiquitin-conjugating enzyme family, is involved in the ubiquitination process where it pairs with Cullin-RING ligases (CRLs) to regulate substrate ubiquitylation and targeted protein degradation [4,6]. This process is crucial for many cellular functions such as controlling an extensive range of eukaryotic biological processes through the formation of Lys48-linked poly-ubiquitin chains [6].
In various cancer-related studies, while not directly from KO/CKO mouse models, the lncRNA UBE2R2-AS1 (antisense RNA 1 of Ube2r2) shows significant associations. In prostate cancer, its high expression is linked to high Gleason score, advanced clinical T stage, lymph node metastasis, and poor prognosis. Knockdown of UBE2R2-AS1 suppresses cell proliferation, migration, invasion, and induces cell cycle arrest and apoptosis [1]. In glioma, decreased UBE2R2-AS1 expression is associated with higher clinical stages, and overexpression inhibits tumor growth in xenograft models [2]. In cervical cancer, UBE2R2-AS1 is downregulated in cancer tissues, and its overexpression suppresses cell proliferation, enhances apoptosis, and reduces cell invasion [3]. In gastric cancer, high UBE2R2-AS1 expression is related to unfavorable clinical pathology and poor prognosis, and it promotes cell proliferation, migration, and invasion [5]. In hepatocellular carcinoma, UBE2R2-AS1 is increased, and its knockdown inhibits growth and metastasis [7]. Also, in gastric cancer, lower UBE2R2-AS1 expression predicts worse drug resistance, and its knockdown enhances cell proliferation and migration [8].
In conclusion, Ube2r2, through its associated lncRNA UBE2R2-AS1, plays a role in the regulation of cell proliferation, apoptosis, invasion, and metastasis in multiple cancer types. These findings from functional studies on UBE2R2-AS1 provide insights into its potential as a diagnostic and therapeutic target in cancer research.
