Overview of Gene Research
Fbxo30, an F-box protein, is a key component of the Skp1-Cullin-F-box complexes, which are ubiquitin E3 ligases. These ligases play a crucial role in the ubiquitin proteasome system, a major protein-degradation pathway in cells. Fbxo30 is involved in multiple biological processes such as cell cycle regulation, chromosome segregation, and tissue development [1,3].
In mouse oocytes, depletion of Fbxo30 causes chromosome compaction, chromosome segregation failure, and arrest at the metaphase of the first meiosis (MI) stage, uncovering its role in oocyte meiosis [1]. In Fbxo30(-/-) female mice, defective mammopoiesis occurs due to Fbxo30's regulation of the bipolar spindle kinesin EG5 through ubiquitinylation, controlling its oscillation during the cell cycle [3]. In clear cell renal cell carcinoma (ccRCC), FBXO30 expression is lower compared to normal adjacent tissues, and it functions as a tumor suppressor by mediating the ubiquitination and degradation of hypoxia-inducible factor-1α (HIF-1α) under normoxia [2]. In neural tube defects, FBXO30 acts as an E3 ubiquitin ligase, targeting retinoic acid receptor γ for ubiquitination and proteasomal degradation, thus regulating BMP signaling [4].
In conclusion, Fbxo30 plays essential roles in various biological processes like oocyte meiosis, mammopoiesis, and in disease conditions such as ccRCC and neural tube defects. Gene-knockout mouse models have been instrumental in revealing these functions, providing insights into the molecular mechanisms underlying these processes and diseases.
