Overview of Gene Research
KATNB1, encoding the non-catalytic regulatory p80 subunit of the katanin microtubule-severing enzyme complex, is crucial for microtubule regulation. Katanin enzymes are involved in multiple cellular processes related to microtubule-based structures. Genetic models, such as gene knockout mouse models, are valuable for studying its functions [2,3,5].
In mouse models, complete loss of KATNB1 from germ cells makes sperm production impossible. It is the master regulator of all katanin enzymatic A-subunits during spermatogenesis, maintaining their abundance. KATNB1 has multiple functions in male meiosis, acrosome formation, sperm tail assembly, regulation of Sertoli and germ cell cytoskeletons during sperm nuclear remodelling, and maintenance of seminiferous epithelium integrity [1].
In zebrafish, katnb1 mutants serve as a new idiopathic scoliosis (IS) model, with Katnb1 having essential roles in motile ciliated lineages [4].
In humans, mutations in KATNB1 are associated with complex cerebral malformations, as patient-derived fibroblasts show defective mitotic spindle formation due to disrupted interactions between mutant KATNB1 and KATNA1 and other microtubule-associated proteins [3].
In conclusion, KATNB1 is essential for microtubule-related functions in various biological processes. Mouse models, especially KO/CKO mouse models, have revealed its key roles in spermatogenesis and provided insights into male infertility. In addition, its mutations in humans are associated with complex cerebral malformations, highlighting its importance in understanding the mechanisms of these diseases.
