Overview of Gene Research
Zbtb3, belonging to the Zinc finger and BTB/POZ domain containing transcription factor family, is involved in multiple biological processes. It plays a role in regulating the ROS detoxification pathway, which is crucial for cancer cell growth [5]. Additionally, it is implicated in chromatin looping-mediated regulation of genes like IRF5, having implications in autoimmune diseases [1,3].

In endometrial cancer, the E3 ubiquitin ligase complex SPOP-CUL3-RBX1 targets ZBTB3 for degradation. EC-associated SPOP mutants are defective in this regulation, leading to ZBTB3 accumulation and promoting endometrial cell proliferation, migration, and invasion. Sonic hedgehog (SHH), a transcriptional target of ZBTB3, is upregulated in this context [2]. In embryonic stem cells (ESCs), lncKdm2b activates Zbtb3, which in turn potentiates ESC self-renewal in a Nanog-dependent manner. Zbtb3 deficiency impairs ESC self-renewal and early embryonic development [4].

In conclusion, Zbtb3 is essential for cancer cell growth through the ROS detoxification system, and is involved in the regulation of endometrial cancer progression and embryonic stem cell self-renewal. The study of Zbtb3 using gene knockout or conditional knockout models in relevant diseases helps to understand its role in these biological processes and offers potential therapeutic targets for cancer and insights into early embryonic development.
