Overview of Gene Research
Slc24a2, also known as solute carrier 24 family member 2, encodes a potassium-dependent sodium-calcium exchanger (NCKX2) that can transport metal ions across the cell membrane [2]. It is involved in maintaining intracellular calcium homeostasis, which is critical for many physiological functions [4].

In the context of diseases, SLC24A2 has been implicated in multiple conditions. In a study on type 2 diabetes (T2D) and cancer, SLC24A2 was identified as a key gene strongly associated with fasting plasma glucose. Overlapped differentially expressed genes between T2D and control, and SLC24A2-high and SLC24A2-low were analyzed, and it was found that SLC24A2 may link T2D and cancer by influencing the ribosome function of islet β cells [1]. In pancreatic ductal adenocarcinoma, a new nonsynonymous single nucleotide variant in SLC24A2 was found, which decreased the stability of the SLC24A2 protein [2].

In neuropathic pain, the expression of SLC24A2 and its encoded protein NCKX2 were downregulated in the posterior horn of the spinal cord in a sciatic nerve chronic constriction injury model. Overexpression of SLC24A2 reduced hyperalgesia and decreased inflammatory cytokines, and miR-135a-5p was shown to regulate SLC24A2 [4]. Additionally, SLC24A2 was among the genes whose expression was affected by valproic acid exposure in human forebrain organoids, and these genes were significantly overlapped with those in autism-spectrum-disorder-related pathways [5].

In colon adenocarcinoma, a prognostic model based on calcium extrusion-related genes including SLC24A2 was constructed, and high-risk scores of these genes were associated with poorer prognosis [6]. Also, SLC24A2 has been associated with skin color variation in Chinese populations [3], and genes near SLC24A2 were associated with the correlation structure of CVD risk factors in a Ghanaian population [7].

In conclusion, Slc24a2 plays essential roles in maintaining intracellular calcium homeostasis and is associated with multiple diseases such as T2D, cancer, neuropathic pain, autism-spectrum disorder, and CVD. Studies on Slc24a2, especially those using in-vivo models like the ones described above, help to understand its biological functions and its implications in disease mechanisms, potentially providing new insights for disease treatment and prevention.
