Osbpl5, or oxysterol binding protein like 5, is involved in lipid metabolism and phosphatidylinositol homeostasis. It may play a role in pathways related to cell survival, such as the AKT survival pathway [2]. The gene is also associated with cholesterol transport [3].
In non-small cell lung cancer (NSCLC), knockdown of Osbpl5 suppressed cell proliferation, migration, invasion, and epithelial-mesenchymal transition (EMT) process. It was identified as a downstream target of miR-526b-3p, and lncRNA LMCD1-AS1 served as a sponge for miR-526b-3p, forming an axis that promotes NSCLC cell progression [1].
In transient cerebral ischemia, conditional knockout mice showed that abolishing the Osbpl5 gene (as part of a circRNA-mediated pathway) significantly inhibited apoptosis and autophagy, suggesting its role in phosphatidylinositol homeostasis between the plasma membrane and endoplasmic reticulum [2].
In ovarian cancer, knockdown of ARL4C, which affects Osbpl5 expression via the Notch-RBP-Jκ-H3K4Me3 pathway, disrupted cholesterol transport from lysosomes to the endoplasmic reticulum and autophagy, attenuating the resistance of ovarian cancer to carboplatin [3].
In summary, Osbpl5 is crucial for lipid-related cellular functions and homeostasis. Gene-knockout and related models have revealed its significance in diseases like NSCLC, transient cerebral ischemia, and ovarian cancer drug resistance, highlighting its potential as a therapeutic target in these disease areas.
