Overview of Gene Research
Itgb6, encoding integrin β6, is a member of the integrin family proteins. It frequently pairs with integrin subunit αv to form integrin αvβ6, a transmembrane heterodimer. Itgb6 determines the expression and availability of αvβ6, playing crucial roles in processes like wound healing. It is also involved in multiple pathways associated with various diseases [1,3,4].
Gene-knockout studies have provided insights into Itgb6's functions. In head-and-neck cancer mouse models, Itgb6 was identified as a key gene mediating differential responses to anti-CD276 treatment. It regulates the expression of CX3CL1, recruiting and activating PF4+ macrophages, thus modulating resistance to anti-CD276 therapy [2]. In cholangiocarcinoma cell lines, ITGB6-knockout led to decreased cell migration, invasion, and down-regulation of PODXL2, suggesting its role in cancer cell motility [5]. In pancreatic cancer, ITGB6 knockdown inhibited cancer cell proliferation and invasion in vitro and in vivo, indicating its association with cancer malignant progression [6].
In conclusion, Itgb6 is essential for regulating integrin αvβ6 and is involved in diverse biological processes. Studies using gene-knockout models have revealed its significance in cancer, such as modulating immune-checkpoint inhibitor resistance, cell migration, invasion, and proliferation. These findings contribute to understanding the role of Itgb6 in disease and may offer potential therapeutic targets.
