Overview of Gene Research
Nrgn, or neurogranin, is a small brain protein expressed in various telencephalic areas. It plays an essential role in synaptic plasticity by regulating the availability of calmodulin (CaM) [4].
In glioma, NRGN acts as a tumor suppressor. LINC00641 can upregulate NRGN by absorbing miR-4262, thus inhibiting glioma cell proliferation [1]. Also, in glioblastoma, lncRNA RBPMS-AS1 promotes NRGN transcription through the miR-301a-3p/CAMTA1 axis, enhancing the radiosensitivity of glioblastoma [5].
In patients with mild traumatic brain injury, serum NRGN levels are significantly higher in those with traumatic intracranial lesions compared to those without, suggesting its potential as a diagnostic biomarker [2]. Moreover, a single-nucleotide polymorphism (rs12807809) in the NRGN gene is associated with abnormal Papez circuit function in schizophrenia patients [3]. Additionally, in the Han Chinese population, polymorphisms in NRGN are associated with schizophrenia, major depressive disorder, and bipolar disorder [8].
In Alzheimer's disease, NRGN levels in neuron-derived extracellular vesicles in plasma are elevated, potentially serving as a biomarker [7]. In adults with obstructive sleep apnea without dementia, higher levels of NRGN in neuron-derived exosomes are associated with cognitive impairment [6].
In conclusion, Nrgn is crucial for synaptic plasticity. Its study in various disease models has revealed its significance in glioma, traumatic brain injury, schizophrenia, mental disorders, Alzheimer's disease, and obstructive sleep apnea-related cognitive impairment. These findings contribute to understanding the disease mechanisms and may offer new diagnostic and therapeutic targets.
