Overview of Gene Research
Jmjd7, or Jumonji-C (JmjC) domain-containing 7, is a 2-oxoglutarate (2OG)-dependent oxygenase. It is involved in multiple functions such as (3S)-lysyl hydroxylation of TRAFAC GTPases like DRG1 and DRG2 [2]. It may also be coupled with the arginine methylation activities of arginine methyltransferases, playing roles in transcription regulation and the biogenesis of membrane-less organelles [1].
In breast cancer cells, depletion of JMJD7 greatly decreases cell proliferation, indicating its role in cancer-related cell growth [4]. In osteoclast differentiation, Jmjd7 acts as a negative regulator. Knockdown of Jmjd7 in RAW 264.7 cells and BMMs enhanced osteoclast differentiation, increased the expression of osteoclast-related genes, and boosted bone resorptive functions [3]. In head and neck squamous cell carcinoma (HNSCC), the read-through fusion gene JMJD7-PLA2G4B promotes cell proliferation and survival. Ablation of JMJD7-PLA2G4B significantly inhibited HNSCC cell proliferation and increased starvation-induced cell death [5].
In conclusion, Jmjd7 plays crucial roles in various biological processes and disease conditions. Its functions in cell proliferation, as seen in breast cancer and HNSCC, and in osteoclast differentiation highlight its significance. These findings from different cellular models contribute to understanding the biological functions of Jmjd7 and its potential as a therapeutic target in related diseases.
