Overview of Gene Research
Marchf6, a transmembrane endoplasmic reticulum E3 ubiquitin ligase, is a crucial regulator in multiple biological processes [1,2,3,4,5,6,7]. It participates in endoplasmic reticulum associated degradation, and many of its substrates are involved in sterol and lipid metabolism [3]. Marchf6 also acts as an NADPH sensor, regulating ferroptosis, a form of cell death caused by excessive iron-dependent lipid peroxidation [1,4,5,8].
POMC-Cre-mediated Marchf6-deficient mice exhibit hyperphagia, reduced energy expenditure, and weight gain [2]. This indicates that Marchf6 is essential for regulating endoplasmic reticulum stress, ferroptosis, and metabolic homeostasis in POMC neurons. In addition, inhibiting ferroptosis rescued the growth of MARCHF6-deficient tumours and the peri-natal lethality of Marchf6-/-mice [1].
In conclusion, Marchf6 is a key regulator in multiple biological pathways, including ferroptosis and metabolic homeostasis. The use of Marchf6 KO/CKO mouse models has revealed its important roles in disease-related processes such as tumour growth, perinatal survival, and metabolic disorders.
