Overview of Gene Research
IDE, short for Insulin-degrading enzyme, is an evolutionarily conserved zinc-dependent metallopeptidase highly expressed in the brain. It is involved in cleaving multiple substrates including insulin and amyloid beta peptides, potentially linking Alzheimer's disease (AD) and type 2 diabetes (T2D) [1].
In IDE knockout (IDE-KO) mice, a specific induction of microgliosis was found in the hippocampus, influencing performance on hippocampal-dependent memory tests in 12-month-old mice. IDE absence also alters microglial responses to environmental signals, impairing the modulation of phenotypic states [1].
In addition, Klf9-/-mice showed impaired cognitive function, and Klf9 was found to stimulate IDE expression and promote Aβ degradation. Silencing IDE blocked this process, and overexpressing IDE in Klf9-/-mice ameliorated cognitive deficits and reduced Aβ content [2].
In conclusion, IDE plays a crucial role in microglial phenotypes, cognitive function, and Aβ degradation, as revealed through IDE-KO mouse models. These findings contribute to understanding the pathogenesis of Alzheimer's disease and suggest that IDE could be a potential therapeutic target for related neurodegenerative diseases [1,2].
