Overview of Gene Research
Zfp503, also known as Nlz2 or Nolz-1, is a zinc-finger containing transcription factor. It is involved in multiple biological processes such as cell fate determination, organ development, and is regulated by various signaling pathways like hedgehog and BMP signaling [4]. Genetic models, especially KO mouse models, have been crucial in understanding its functions.
In eye development, Zfp503 KO mice exhibit uveal coloboma due to failure of optic fissure closure and abnormal RPE differentiation. Altered mRNA levels and expression patterns of key eye-development transcription factors were observed, leading to reduced melanin pigmentation and RPE differentiation into a neural retina-like lineage [1].
In striatal development, D1 MSNs in Zfp503-deficient mice switch to D2 MSNs, and Zfp503 affects progenitor cell differentiation in the lateral ganglionic eminence by regulating Sp8/9 expression [2].
In the developing brain, CD93 activates Zfp503 transcription through β-Catenin, and Zfp503, as a transcriptional repressor, inhibits astrogenesis by binding to the Gfap promoter with Grg5 [3].
In conclusion, Zfp503 plays essential roles in eye development, neural cell fate determination, and astrogenesis. Studies using KO mouse models have revealed its significance in these processes, providing insights into related congenital eye malformations and neural development-associated disorders [1,2,3].
