Overview of Gene Research
Gsta3, glutathione S-transferase alpha 3, is a member of the glutathione S-transferase family with antioxidative function. It is involved in the PI3K-Keap1/Nrf2 pathway, which plays a role in regulating oxidative stress and has significance in various biological processes and disease resistance [1].
In renal interstitial fibrosis, Gsta3 knockdown in NRK-52E cells led to increased production of reactive oxygen species and tubular epithelial-mesenchymal transition, while overexpressing Gsta3 ameliorated these effects, indicating its protective role against tubular EMT in renal fibrosis [2].
In Gsta3 knockout mice, hepatic aflatoxin B1-N(7)-guanine levels were elevated 120-fold compared with wild-type, and the inability to rescue these mice from aflatoxin genotoxicity through the Nrf2 transcriptional program shows that Gsta3 is unilaterally responsible for the detoxication of aflatoxin in mice [3].
In cutaneous squamous cell carcinoma (cSCC), Gsta3 was downregulated. Overexpression of Gsta3 induced a less malignant phenotype, while its silencing led to more malignant phenotypes, and Gsta3 could inhibit cSCC progression through suppressing the TGF-β/Smad and HIF-1α signalings [4].
In conclusion, Gsta3 plays crucial roles in antioxidation, renal interstitial fibrosis prevention, aflatoxin detoxication, and inhibition of cSCC progression. Gene knockout mouse models have significantly contributed to revealing its functions in these disease-related biological processes, providing potential therapeutic targets for relevant diseases.
