Overview of Gene Research
Lyve1, or lymphatic vessel endothelial hyaluronan receptor-1, is a key marker and functional molecule involved in multiple biological processes. It is associated with cells like macrophages and endothelial cells. In macrophages, it is linked to functions such as tissue homeostasis, immune regulation, and extracellular matrix remodeling. In endothelial cells, it may play a role in revascularization and sinusoid reconstruction [2,3,4]. Genetic models, especially mouse models, are valuable for studying Lyve1's functions.
In mouse models, studies have revealed the heterogeneity of Lyve1+ perivascular macrophages (pvMs) in the brain. Using reporter mouse models, a CX3CR1-pvM population sharing phagocytic functions with Lyve1+F4/80+CD206+CX3CR1+ pvMs was identified. Also, brain parenchyma vasculature mostly hosts Lyve1+MHCII-pvMs with low to intermediate CD45 expression. Lineage tracing models suggest a bone marrow-independent replenishment of all Lyve1+ pvMs in the adult mouse brain [1]. In the skin, macrophage-mediated HA clearance requires Lyve-1 sensing, and loss of CCR2-HDMs (which rely on Lyve-1 for HA clearance) results in HA accumulation and protection against Staphylococcus aureus-induced cellulitis [4].
In conclusion, Lyve1 is crucial for macrophage-related functions in the brain and skin. Mouse models, including lineage tracing and cell-specific depletion models, have been instrumental in revealing these functions. In the context of diseases, understanding Lyve1's role in macrophage-mediated processes such as in stroke-related changes in pvMs and in skin infections can potentially lead to new therapeutic strategies [1,4].
