Overview of Gene Research
Coro1a, short for Coronin 1A, is an actin-binding protein that plays crucial roles in multiple biological processes. It is involved in cytoskeleton remodeling, which is essential for various cellular functions such as cell movement, endocytosis, and cell-cell communication. Additionally, it participates in pathways related to steroidogenesis, neuronal morphogenesis, and autophagy [1,2,4].
In studies related to cadmium (Cd) exposure, Cd-treated male offspring showed up-regulation of Coro1a in testis, leading to F-actin disassembly, reduced LDL uptake, and decreased progesterone production. Silencing Coro1a rescued these effects, highlighting its role in Cd-induced steroid hormone deficiency [1]. In neuronal morphogenesis, Coro1A localizes to growth cones and filopodial structures and is required for netrin-dependent axon turning, branching, and corpus callosum development, collaborating with TRIM67 [2]. In triple-negative breast cancer (TNBC), CORO1A is crucial for the disease progression, and its degradation by a molecular glue degrader via TRIM4 exerted antitumor effects [3]. In non-alcoholic steatohepatitis (NASH), CREBH inhibits Coro1a expression, and overexpression of Coro1a in liver cells inhibits autophagic flux and elevates inflammatory cytokine levels [4].
In conclusion, Coro1a is essential for cytoskeleton-related functions, steroidogenesis, neuronal development, and autophagy-related processes. Its study through gene-based models has provided insights into diseases like Cd-induced steroid hormone deficiency, TNBC, and NASH, highlighting its potential as a therapeutic target in these disease areas.
