Pcdha11, encoding protocadherin alpha 11, is likely involved in cell-cell adhesion and communication, which are crucial for normal tissue development and function. It may be associated with multiple signaling pathways such as the AKT/mTOR, Wnt/β-catenin, and JAK/STAT pathways, given its impact on cellular functions in cancer cells [1].
In a study on gastric cancer, AmNA-modified anti-PCDHA11 antisense oligonucleotides (ASOs) significantly reduced the proliferation of gastric cancer cells and other solid tumors. Pcdha11-deficient mice demonstrated normal reproductive, organ, and motor functions, yet the ASOs disrupted the AKT/mTOR, Wnt/β-catenin, and JAK/STAT signaling pathways in cancer cells, inhibiting tumor growth in mouse models of peritoneal metastasis, systemic metastasis, and established subcutaneous tumors [1]. In lung squamous cell carcinoma (LUSC), silencing PCDHA11 enhanced the anticancer effects of cisplatin, while its overexpression weakened these effects, suggesting its role in chemoresistance [2].
In summary, Pcdha11 seems to play a role in the proliferation and chemosensitivity of cancer cells, especially in gastric and lung squamous cell carcinomas. Studies using Pcdha11-deficient mouse models and manipulation of PCDHA11 expression in cancer cells have provided insights into its function in tumor-related biological processes, which may contribute to the development of new cancer treatment strategies [1,2].
