Overview of Gene Research
Csf2rb2, also known as Il3r, is a β chain cytokine receptor. It is involved in myeloid cytokine signaling pathways and plays a role in hematopoiesis, which is crucial for the production and regulation of blood cells [1,2,3]. Genetic models, such as gene knockout mouse models, can be used to study its functions.
In a study, conditional disruption of RBP-J (a transcription factor of canonical Notch signaling) increased irradiation sensitivity in mice. Activation of Notch signaling with mD1R promoted bone marrow recovery after irradiation, and this was associated with an increase in myeloid progenitors and monocytes. Mechanistically, mD1R increased the proliferation and reduced the apoptosis of myeloid cells, and it was found that Csf2rb2 was a downstream molecule of Notch signaling in hematopoietic cells, with mD1R improving hematopoietic recovery through up-regulation of Csf2rb2 expression [1]. In another study, deletion of Rcor1 in mice led to profound anemia and arrest of definitive erythroid cells at a specific stage. The Csf2rb gene (coding for a receptor implicated in myeloid cytokine signaling) was a direct target for both Rcor1 and the transcription repressor Gfi1b in erythroid cells. In the absence of Rcor1, the Csf2rb gene was highly induced, and Rcor1 -/- progenitors exhibited CSF2-dependent phospho-Stat5 hypersensitivity [2].
In conclusion, Csf2rb2 is essential for hematopoiesis, influencing the proliferation and apoptosis of myeloid cells and the differentiation of erythroid cells. The study of Csf2rb2 in KO mouse models reveals its importance in irradiation-and drug-induced bone marrow suppression, providing potential insights for the treatment of related hematopoietic disorders [1,2].
