Overview of Gene Research
Asb13, an ankyrin repeat and suppressor of cytokine signaling (SOCS) box-containing E3 ubiquitin ligase, is evolutionarily conserved and may be involved in regulating compartment size, such as in the brain and muscle [2]. It is also implicated in canonical Notch signaling and mitochondrial function [2].
In breast cancer, ASB13 knockout in cells promoted cell migration and decreased F-actin polymerization, while its overexpression suppressed lung metastasis. ASB13 acts by promoting the ubiquitination and degradation of transcription factor SNAI2, which in turn relieves the transcriptional repression of YAP, a suppressor of breast cancer progression. Clinical data shows ASB13 expression is positively correlated with improved overall survival in breast cancer patients [1]. In young women with breast cancer, CNA mutations in ASB13 were associated with higher gene expression and a worse survival outcome [3]. In acute myocardial infarction treated with extracorporeal membrane oxygenation, ASB13 was identified as a prognostic marker, and its co-expressed proteins are related to post-translational protein modification, cullin-RING ubiquitin ligase complex, and ubiquitin-mediated proteolysis [4].
In summary, Asb13 plays significant roles in breast cancer metastasis and prognosis, as well as in the context of acute myocardial infarction treatment. Gene knockout-based research, like in breast cancer cells, has been crucial in revealing its role in suppressing breast cancer metastasis by modulating key proteins and pathways [1,3].
