Overview of Gene Research
Tnks1bp1, originally identified as an interaction protein of tankyrase 1, belongs to the poly(ADP-ribose) polymerase (PARP) superfamily. It is involved in multiple biological processes. In DNA repair, it participates in the DNA double-strand break repair machinery. It also has a role in regulating the actin cytoskeleton and is associated with pathways related to cell invasion and autophagy, thus being of great importance in cancer-related biological events [3,4].
In hepatocellular carcinoma (HCC), knocking down Tnks1bp1 synergizes with anti-PD-L1 treatment. Tnks1bp1 interacts with TRIM21 to mediate the ubiquitination of CNOT4, promoting HCC progression and immune evasion. In radiation-induced lung injury, Tnks1bp1 knockout (KO) mice demonstrate prevention of IR-induced lung tissue senescence and injury as Tnks1bp1 up-regulation causes AECII senescence [1,2].
In conclusion, Tnks1bp1 is crucial in DNA repair, cell invasion, autophagy, and senescence processes. The use of Tnks1bp1 KO mouse models has revealed its role in HCC and radiation-induced lung injury, providing potential therapeutic targets for these diseases.
